PFKFB3 blockade reduces angiogenesis. (A) Current model of vessel sprouting, relying on a migratory tip cell with filopodia that competes for the tip position. Proliferating stalk cells elongate the sprout. Once perfused, ECs turn into quiescent phalanx cells. (B) Blocking PFKFB3 results in impaired proliferation and migration of endothelial stalk and tip cells, respectively, and impedes the dynamic overtaking of ECs typically seen during physiological angiogenesis, all resulting in impaired vessel sprouting.
